I don't have to delve too far into my gaming past to find my saddest scene--it's from one of the last games I've played.  In Assassins Creed: Brotherhood, you have access to optional memories involving Ezio's brief flame from II, Cristina.  They all deal with him crossing paths with Cristina while on his path for revenge and his rise as an assassin.  In this one, the second memory, Ezio revisits the day his brothers were killed.

I've never had a problem with playing a game before, but as I had to take the bodies of Ezio's dead family members one by one and carry them to a nearby boat so he could properly bury them, I'll admit I had issues.  A couple of times I had to walk away and come back to it.  I'm pretty sure I cried.  It was just so powerful, so incredibly sad, that I couldn't do anything else.  It was the hardest thing I've ever had to do in a game emotionally, and the saddest scene I've ever witnessed.
